Reaction products for the enhanced chemiluminescence reaction studied by HPLC method

The kinetic parameters for individual and co-oxidation of N,N-dimethylaminoluminol (DAL) and p-iodophenol (PIP)by hydrogen peroxide in the presence of alkaline izoenzymes of horseradish peroxidase (HRPB) were studied.The rate constants for DAL oxidation by compound I and compound II of HRP were determined by means of stop-flow technique. DAL was oxidized by hydrogen peroxide in the presence of HRPB by approximately ten times slowly than luminol. The main products of DAL oxidation were determined by HPLC as follows: AP is N,N-dimethylaminophtalate acid, B - 5-dimethylaminoformylbenzen acid, C - 4-dimethylamino-formylbenzen acid. B and C are the products of diazoquinon hydrolysis and they are dark products. The ratio between light and dark products varies at co-oxidation of DAL and PIP catalyzed by HRPB, as B was not detected in the course of co-oxidation, either its content was less in a comparison with individual peroxidation of DAL. The enhancer was found to increase the luminescence quantum yield to DAL due to the increase of superoxide-ion steady-state concentration as a result of interaction of PIP radicals with dissolved oxygen or hydrogen peroxide.[Poster: egorov.alexey.20132]
