Characterization of acridinium-9-carboxamide-antibody conjugates by mass spectrometry

A label covalently attached to an antibody can provide it with the ability to generate a signal. Chemiluminescent bioconjugates labeled with acridinium salts, are used in ultra sensitive immunoassays. Determination of the number and distribution of bound labels is an essential aspect of characterizing such conjugates. Traditional methods such as UV-VIS or fluorescence are problematic. These methods do not differentiate between covalently bound, non-specifically bound, and unbound acridinium labels.
We present a direct method for determining the extent and distribution of acridinium-modified antibodies, as well as the regiospecificity of the conjugation (Fab/Fc), using electrospray ionization mass spectrometry. Together with quantification of free chemiluminescent material, the method provides a comprehensive approach to the characterization of chemiluminescent bioconjugates.
