Molecular genetic analysis of circadian clocks

Our laboratory is interested in understanding the interactions between phototransduction pathways and the circadian clock in controlling plant growth and development. Using cab-luciferase transgenic lines as bioluminescent markers, we are undertaking several different approaches that will be discussed:

1) Identification and characterization of novel circadian rhythm mutants by screening for aberrant period length (toc) mutants. Two of these are currently being characterized in detail.
2) Genetic analysis of photoreceptor-clock interactions using a wide range of photoreceptor/phototransduction mutant backgrounds.
3) Genetic analysis of the role of the circadian clock in the photoperiodic
control of flowering time.
4) Analysis of DNA-protein interactions mediating clock- and phytochrome-controlled gene transcription.
5) Identification of novel markers for clock regulation at different stages of development and in different tissues.

We will discuss how these approaches have revealed a central role for the circadian clock in controlling a wide range of plant metabolic and developmental processes.
