Method for real-time detection of terminaltransferase activity

A simple bioluminomertic assay for detection of terminaltranferase activity has been developed. The technique takes advantages of a two-coupled enzyme mixture to monitor terminaltranferase activity. The PPi released during the terminaltransferase catalyzed polymerization is converted to ATP by ATP sulfurylase and the ATP production is continuously monitored by firefly luciferase. The assay is sensitive and yields linear responses between 0.002 to 100 mU of terminaltransferase. The detection limit was found to be 20 microunits of terminaltransferase from calf thymus. The effect of different nucleotides and oligonucleotides on the terminaltransferase activity was studied.
